Findings reported in the Journal of Dairy Science show promise for treatment of gastric cancer



Philadelphia, PA, November 7, 2013 New research from a team of researchers in Taiwan indicates that a peptide fragment derived from cow's milk, known as lactoferricin B25 (LFcinB25), exhibited potent anticancer capability against human stomach cancer cell cultures. The findings, published in the Journal of Dairy Science, provide support for future use of LFcinB25 as a potential therapeutic agent for gastric cancer.


"Gastric cancer is one of the most common causes of cancer-related mortality worldwide, especially in Asian countries," says Wei-Jung Chen, PhD, of the Department of Biotechnology and Animal Science of National Ilan University, Taiwan Republic of China. "In general, the main curative therapies for gastric cancer are surgery and chemotherapy, which are generally only successful if the cancer is diagnosed at an early stage. Novel treatment strategies to improve prognosis are urgently needed."


Investigators evaluated the effects of three peptide fragments derived from lactoferricin B, a peptide in milk that has antimicrobial properties. Only one of the fragments, LFcinB25 reduced the survival of human AGS (Gastric Adenocarcinoma) cells in a dose-dependent and time-dependent manner.


Under a microscope the investigators could see that after an hour of exposure to the gastric cancer cells, LFcinB25 migrated to the cell membrane of the AGS cells, and within 24 hours the cancer cells had shrunken in size and lost their ability to adhere to surfaces. In the early stages of exposure, LFcinB25 reduced cell viability through both apoptosis (programmed cell death) and autophagy (degradation and recycling of obsolete or damaged cell parts). At later stages, apoptosis appeared to dominate, possibly through caspase-dependent mechanisms, and autophagy waned.


"This is the first report describing interplay between apoptosis and autophagy in LFcinB-induced cell death of cancer cells," says Dr. Chen.


The research also suggested a target, Beclin-1, which may enhance LFcinB25's cytotoxic action. Beclin-1 is a protein in humans that plays a central role in autophagy, tumor growth, and degeneration of neurons. In this study, the investigators found that cleaved beclin-1 increased in a time-dependent manner after LFcinB25-exposure, suggesting to the authors a new approach in drug development that may boost the anticancer effects of LFcinB25.


"Optimization of LFcinB using various strategies to enhance further selectivity is expected to yield novel anticancer drugs with chemotherapeutic potential for the treatment of gastric cancer," concludes Dr. Chen.

NEW YORK, NY (Nov. 7, 2013) A Columbia University Medical Center-led research team has clinically validated a new method for predicting time to full-time care, nursing home residence, or death for patients with Alzheimer's disease. The method, which uses data gathered from a single patient visit, is based on a complex model of Alzheimer's disease progression that the researchers developed by consecutively following two sets of Alzheimer's patients for 10 years each. The results were published online ahead of print in the Journal of Alzheimer's Disease.


"Predicting Alzheimer's progression has been a challenge because the disease varies significantly from one person to anothertwo Alzheimer's patients may both appear to have mild forms of the disease, yet one may progress rapidly, while the other progresses much more slowly," said senior author Yaakov Stern, PhD, professor of neuropsychology (in neurology, psychiatry, and psychology and in the Taub Institute for Research on Alzheimer's Disease and the Aging Brain and the Gertrude H. Sergievsky Center) at CUMC. "Our method enables clinicians to predict the disease path with great specificity."


"Until now, some methods of predicting the course of Alzheimer's have required data not obtained in routine clinical practice, such as specific neuropsychological or other measurements, and have been relatively inaccurate. This method is more practical for routine use," said Nikolaos Scarmeas, MD, a study co-author and associate professor of neurology, in the Taub Institute and the Sergievsky Center. "It may become a valuable tool for both physicians and patients' families."


The new method also may be used in clinical trialsto ensure that patient cohorts are balanced between those with faster-progressing Alzheimer's and those with slower-progressing diseaseand by health economists to predict the economic impact of Alzheimer's disease.


The prediction method is based on a Longitudinal Grade of Membership (L-GoM) model, developed by a research team also led by Dr. Stern and published in 2010.


The L-GoM includes 16 sets of variables, such as ability to participate in routine day-to-day activities; mental status; motor skills; estimated time of symptom onset; and duration of tremor, rigidity, or other neurological symptoms. It also includes data obtained postmortem (time and cause of death).



"The benefit of the L-GoM model is that it takes into account the complexity of Alzheimer's disease. Patients don't typically fall neatly into mild, moderate, or severe disease categories. For example, a patient may be able to live independently yet have hallucinations or behavioral outbursts," said Dr. Stern, who also directs the Cognitive Neuroscience Division at CUMC. "Our method is flexible enough to handle missing data. Not all 16 variables are needed for accurate predictionsjust as many as are available."


Results can be presented as expected time to a particular outcome. Two 68-year-old Alzheimer's patients, for example, had similar mental status scores (one a mini-mental status score (mMMS) of 38/54, the other of 39/54) at initial visit. The first patient was more dependent on his caregiver and had psychiatric symptoms (delusions). These and other subtle differences in the initial presentation of the two patients resulted in different predictions of time until death. The method accurately predicted that the first patient would die within three years, while the other would survive more than 10 years.


"In addition to time to nursing home residence or death, our method can be used to predict time to assisted living or other levels of care, such as needing help with eating or dressing, or time to incontinence," said first author Ray Razlighi, PhD, assistant professor of neurology at CUMC and adjunct assistant professor of biomedical engineering at Columbia University.


Development of the method began in 1989, when Dr. Stern received a grant from the National Institutes of Health to begin the Predictors of Severity in Alzheimer's Disease study. "The fact that work on this prediction method began nearly 25 years ago underlines the difficulties of studying Alzheimer's disease," said Richard Mayeux, MD, MS, neurology chair, the Gertrude H. Sergievsky Professor of Neurology, Psychiatry and Epidemiology and co-director of the Taub Institute and the Sergievsky Center.


Dr. Stern and colleagues at Massachusetts General Hospital and Johns Hopkins first followed 252 non-familial Alzheimer's patients every six months for 10 years. Eric Stallard, an actuary at Duke and a co-author of the paper, used the resultant data to create an L-GoM model of Alzheimer's progression. They published their results in 2010 in Medical Decision Making. The researchers then followed a separate group of 254 patients and used data from only a single patient visit to predict outcomes for this group.


Dr. Stern and his team are now developing a computer program that would allow clinicians to input the variables and receive a report. They expect the program to become available within the next two years. Eventually, such a program might be incorporated into electronic health records. "At our Alzheimer's center, patients are already filling out much of their clinical information electronically," said Dr. Stern.


The researchers are also testing the method with a third cohort. While the first two sets of patients were primarily white, educated, and of high socioeconomic status, the new cohort follows a diverse group of participants from CUMC's Washington Heights-Inwood Columbia Aging Project (WHICAP), an ongoing, community-based study of aging and dementia comprising elderly, urban-dwelling residents. Because participants may be dementia-free when they join the study, the researchers are able to capture the age of dementia onset and track symptom development over time.

The paper is titled, "A New Algorithm for Predicting Time to Disease Endpoints in Alzheimer's Disease Patients." The method's formula is detailed in supplementary material for the paper. The other contributors are: Anatoliy I. Yashin (Duke); Jason Brandt and Marilyn Albert (Johns Hopkins); Deborah Blacker (Massachusetts General Hospital/Harvard); and Bruce Kinosian (Philadelphia VA Medical Center).

After offering birthday wishes to friend Brittny Gastineau, Kim Kardashian inadvertently raised suspicions that she is linked to the Illuminati, a conspiratorial group that theorists believe controls the world.


On Wednesday (November 7), the "Keeping up with the Kardashians" starlet posted an Instagram message to Brittny that read, “Happy Birthday to my best friend @brittgastineau We’ve shared soooo many memories over the past years! I love you! #RideorDie.” However the confusion surrounds the nature of the photo attached to the post. The snapshot features a collage of the two besties, except it showcases them within the monogram of the iconic pyramid with an eye in the center.


After hearing the feedback from followers linking her to the group, the 33-year-old posted a response the following day, writing, "I posted a IG collage for my bff @BrittGastineau & people say its the illuminate!"


Clearly confused, Kim also wrote, "What is the illuminate? A religion?… I’m a Christian. A cult?… not into that sorry! It had an eye on it, which reminded me of Britt bc her company ‘eye on glam’ & reminds me of against evil eye bracelets we wear. Maria Menounos Shares Healthy Holiday Tips: Watch Here!

AP sources: Kerry to join Iran nuclear talks


GENEVA (AP) — Officials say U.S. Secretary of State John Kerry will fly to Geneva on Friday to participate in nuclear negotiations with Iran and other major powers.

The officials say Kerry will travel to the talks after a brief stop in Israel, where he will hold a third meeting with Israeli Prime Minister Benjamin Netanyahu. Israel's intense interest in Iran's nuclear ambitions is a likely topic between Netanyahu and Kerry as well as Israeli-Palestinian peace.

Iran's plan to cap some of the country's atomic activities in exchange for selective relief from crippling economic sanctions has been accepted by six world powers, the country's chief nuclear negotiator said Thursday.

Kerry's last-minute decision to join the talks suggests a deal could be imminent.

The officials spoke on condition of anonymity because Kerry has not been formally invited by the Europeans to join the talks.

Researchers suggest plan to address hypoxia in Gulf of Mexico

URBANA, Ill. Despite a 12-year action plan calling for reducing the hypoxia zone in the Gulf of Mexico, little progress has been made, and there is no evidence that nutrient loading to the Gulf has decreased during this period. University of Illinois researchers have identified some of the biophysical and social barriers to progress and propose a way forward.


"We are suggesting that a partnership of researchers work closely with farmers to develop the suite of practices that are needed to reduce nutrient losses from agricultural fields," said U of I biogeochemist Mark David who has been studying nitrate loss since 1993. "Working with farmers is essential to develop realistic practices on real-world farmswhere the constraints that influence management are presentto document the effectiveness, and to communicate the environmental and socioeconomic results regionally."


"Biophysical and Social Barriers Restrict Water Quality Improvements in the Mississippi River Basin" was published in the Nov. 5 issue of Environmental Science and Technology. The opinion piece was authored by researchers Mark B. David, Courtney G. Flint, Gregory F. McIsaac, Lowell E. Gentry, Mallory K. Dolan, and George F. Czapar.


David said that the Gulf of Mexico hypoxic zone that was measured in July 2013 was 5,800 square miles (nearly the size of Connecticut), the result of riverine losses of nitrate and total phosphorus from the Mississippi River Basin. The goal of the 2008 action plan is to reduce the zone to a five-year running average of 2,000 square miles by 2015 and calls for a 45 percent reduction in total nitrogen and phosphorus, but these goals have not been met.


"Much of the nitrate that leads to the hypoxic zone formation is lost from millions of acres of fields across the upper Midwest, where drainage has been accelerated by a variety of agricultural practices," David said. "Many flat agricultural fields are artificially drained with perforated plastic tubing or older clay drainage tiles to allow timely field work and enhance crop growth. There are now tens of millions of acres of tile-drained fields with large losses of nitrate, even with the recommended best management practices being followed," he said.


According to the researchers, the combination of expanded and patterned tile drainage, increased fertilizer use due to more corn production, and more frequent high-intensity precipitation events all contribute to greater losses of nutrients and therefore a large hypoxic zone. This occurs even though nutrient balances (inputs minus outputs) have generally improved across the upper Midwest.


The USDA Natural Resources Conservation Service (NRCS) promotes and provides technical information on a wide array of techniques that can be used to reduce nutrient losses, including fertilizer rate, timing and placement; cover crops; nitrification inhibiters; water table management; tile bioreactors; constructed wetlands; buffer strips; and conversion of row crops to CRP or perennial crops. David said that unfortunately, few of these methods are used on tile-drained fields because they impose substantial costs and/or risks on the producer without increasing crop production.


"For example, end-of-pipe practices such as tile bioreactors or constructed wetlands have substantial construction costs, require land to be taken out of production, and provide no production benefit to the producer," David said.


The researchers noted that important constraints are in the socioeconomic realm and relate to factors influencing adoption of farm conservation practices.


"Producers view themselves as stewards who care for the land, but they need to make a living from it," said rural sociologist Courtney Flint. "Not only can they not see the loss of nutrients, they are disconnected physically from the downstream effects.
Stewardship objectives may be strong, but they can be trumped or complicated by other economic, social, and environmental drivers.


"Additionally, there is a growing sense among farmers that policy makers are too far removed from the realities of farming," Flint added. "This leads to an ever-widening trust gap that is a major barrier to effective collaboration and policy development for water-quality improvement in the Mississippi River Basin and beyond."


The researchers believe that having farmers actively participate with researchers to develop realistic suites of practices could find widespread regional acceptance, but they realize that real-world, on-farm longitudinal studies of nutrient loss reduction practices will require considerable funding for cost-sharing practice development and implementation.

Apple fixes Gmail bug in latest Mail update for Mavericks


Mavericks may have brought a number of notable improvements to OS X, but Gmail integration was certainly not one of them. Users of Google's email service might've encountered an unpleasant surprise when they upgraded their operating systems to Apple's latest, as the changes to Apple Mail appeared to be incompatible with how Gmail worked. For background, Gmail apparently has a rather unorthodox method of using IMAP, so you needed to set up a roundabout method that prevents Mail from downloading every single message from Gmail twice (Basically, you had to tweak your Gmail settings so that "All Mail" is hidden from email programs).


Mavericks, however, broke that workaround, so users of both Gmail and Apple Mail ended up suffering from bloated inboxes. There were other issues too, like the inability to organize mailboxes and unread message numbers that are totally off. Thankfully, however, Apple has just released an update that fixes all that. The release states that it "fixes an issue that prevents deleting, moving, and archiving messages for users with custom Gmail settings" and "addresses an issue that may cause unread counts to be inaccurate." So for those who've bitten the Mavericks bug and want a Mail app that plays nicer with Google's email, you should definitely hit that update button right about now.


Update: Also, remember that keyboard and trackpad glitch on the 13" MacBook Pro? Well, Apple's apparently fixed that in an update as well. Get to downloading, everyone!

Mary-Kate & Ashley Olsen: We Never Felt Like Actresses

They’ve been in the public eye since the young age of nine months, but Mary-Kate and Ashley Olsen reveal that they never really felt like it was right.


Though the uber-famous twins gained extraordinary wealth and success, the 27-year-old told Women’s Wear Daily that they are finished with Hollywood and ready to focus primarily on fashion.


“We were always hard workers as kids,” Mary-Kate tells WWD. “It’s just the way we are, the way we were raised. Our work ethic was everything, and that never left us. We like to work hard, and we like to try to do everything 100 percent. In fact, it’s actually almost impossible for us not to. Sometimes you succeed and sometimes not so much, but it’s learning.”


Mary-Kate and Ashley run Dualstar Entertainment Group, which manufactures various items including fragrances, in addition to their clothing brands like The Row and Elizabeth and James.


They’ve found what they are comfortable doing now, and admit that they never felt comfortable on set.


“With what we were doing in business when we were younger, I don’t think it ever felt like we were actresses — because we spent so much more of our time not in front of the cameras, building a brand," Mary-Kate continued.


“I always looked at myself, even as a kid, as a businesswoman,” Ashley added.


“We feel so fortunate to have each other, to have a dialogue,” Ashley explained. “Communication is key; it’s the most important thing in life. And Mary-Kate and I get to communicate all day long, on all sorts of subjects. It gets us to a more educated, thoughtful place, because we often come to things from different directions — although we want to get to the same space and have the same goal and vision. But that conversation is what gets us there.”

Toronto mayor, hammered, appears in another strange video

TORONTO (Reuters) - Just days after Toronto Mayor Rob Ford apologized for smoking crack cocaine, he admitted on Thursday he was "extremely, extremely inebriated" in a short expletive-laden video posted online.

The news hit as a campaign by some city councillors to ask the provincial government to remove Ford gathered steam.

The blurry, 80-second clip, posted on the Toronto Star's website and shown on Canadian television, shows a clearly agitated Ford ranting and pounding his hands together, while at least one other person seems to goad him on.

The Toronto Star said it had paid for the video, and said the context of the video was not clear.

"He dies or I die," Ford says in the clip, which also refers to something happening "in that ring." He adds: "I need fucking 10 minutes to make sure he's dead. It'll be over in 5 minutes."

Minutes after the video was posted, Ford emerged from his City Hall office to apologize.

"All I can say is - again - I've made mistakes. All I can do is reassure the people that ... I just wanted to come out and tell you I saw the video, it's extremely embarrassing," Ford said. "Obviously, I was extremely, extremely inebriated."

The mayor did not say who he was talking about in the video.

On Tuesday, Ford made international headlines when he admitted he had smoked crack cocaine, "probably in one of my drunken stupors." He apologized and promised it would never happen again.

For months, the mayor had been dodging questions about reports by media blog Gawker and the Toronto Star that he had been caught on tape using the drug.

Toronto Police Chief Bill Blair said last week the force had obtained a video "consistent" with those reports, without describing its contents in detail.

Ford has refused to resign, and vowed to run for re-election next October. It is difficult to force the mayor of Toronto out of office, unless he or she violates election or conflict of interest rules, or goes to prison. There are no recall elections of the sort that take place in the United States.

But Toronto's city council may vote next week to ask the provincial government to remove Ford from his job.

Councillor Denzil Minnan-Wong, a former Ford supporter, drafted that motion. He told local media on Thursday that he does not believe the province would act without the support of a "large majority" of council.

Even some who want Ford ousted may balk. Councillor James Pasternak warned that the motion could set a "dangerous and risky precedent," but did not rule out supporting it.

"I'm hoping for a dignified exit before then, and that way we can spare any more harm to the city," he told reporters on Thursday.

At one point in the video posted on Thursday, an unknown voice says "Mike Tyson," presumably referring to the boxer. Earlier this year Ford arm-wrestled Hulk Hogan, the professional wrestler, at a media event.

The Toronto Sun, which posted a few seconds from the video, said a source close to the mayor had confirmed it showed Ford, but said it "looks a lot worse than it really is" because the mayor "sometimes goes off on tangents. (Reporting by Allison Martell; editing by Janet Guttsman and Jackie Frank)
